The supplement industry has a reliable formula for selling NMN: NMN raises NAD+, NAD+ declines with age, buy this capsule. What the label never addresses is the chemistry between swallowing that capsule and NAD+ appearing in your mitochondria.
That gap is where 90% of most NMN doses disappear.
I hold an M.Sc. in Organic Chemistry. I’ve spent the last year analyzing longevity supplement formulations through a pharmacokinetics lens rather than a marketing one. What follows is a clinical breakdown of why NMN bioavailability is the central variable the industry least wants you to understand.
The Pathway NMN Is Supposed to Travel
Before the failure modes, here is what intact NMN is trying to do:
Oral NMN → intestinal absorption via Slc12a8 transporter → intracellular NMNAT enzyme → NAD+
Nicotinamide mononucleotide adenylyltransferase (NMNAT) converts intact NMN into NAD+ in a single enzymatic step. This is the most direct biosynthetic route to NAD+ available in supplement form. One molecule in, one pathway, one output.
Everything that happens between the capsule dissolving and intracellular delivery is attrition. There are three layers of it, and they are cumulative.
Layer 1: Gastric Hydrolysis
NMN (molecular formula C₁₁H₁₅N₂O₈P, MW 334.22 g/mol) contains a phosphate ester bond that is thermodynamically unstable at stomach pH of 1.5–3.5. Standard vegetarian capsules dissolve within 5–10 minutes of gastric contact, exposing raw NMN powder to full acid for the entire gastric residence time — typically 30–90 minutes.
Acid-catalyzed hydrolysis cleaves the phosphate from the ribose, converting NMN first to nicotinamide riboside (NR), and with sustained exposure, further to plain nicotinamide — Vitamin B3, available for under $5.
This happens to every standard capsule product. Premium and generic alike.
The solution: Enteric coating (pH-triggered intestinal release) or liposomal encapsulation (phospholipid shell resists gastric acid). Only liposomal addresses all three layers simultaneously.
Layer 2: CD73 Dephosphorylation
Even NMN that survives gastric transit faces a second interception at the intestinal brush border. CD73 — formally ecto-5′-nucleotidase — is an ectoenzyme expressed on intestinal epithelial cell surfaces. It dephosphorylates NMN to nicotinamide riboside (NR) before the NMN-specific Slc12a8 transporter can absorb it.
Grozio et al. (2019, Nature Metabolism, PMID: 31263207) identified Slc12a8 as the dedicated NMN transporter with highest expression in the jejunum and ileum. CD73 works upstream of it, at the brush border. The cell must then re-phosphorylate NR back to NMN via NRK enzymes before NMNAT can act — adding enzymatic steps and conversion inefficiency to every milligram that took this route.
Sublingual advocates point to oral mucosal absorption as a CD73 bypass. This is mechanistically correct — but approximately 70% of any sublingual dose is swallowed and faces the full intestinal pathway anyway.
Liposomal bypass: Endocytic uptake delivers the phospholipid vesicle whole into intestinal epithelial cells. CD73 sits on the outside of those cells. Intracellular NMN is simply never accessible to it.
Layer 3: Gut Microbial Deamidation
Research published in Science Advances demonstrated that gut microbiota deamidate a significant fraction of orally administered NMN before systemic absorption. The product is nicotinic acid (NA) — a structurally distinct NAD+ precursor that enters the Preiss-Handler pathway rather than the direct NMNAT salvage route.
The Preiss-Handler pathway requires three enzymatic steps to reach NAD+ versus one for intact NMN. Lower yield per milligram of original dose. More energetically costly. And critically — the magnitude of microbial deamidation varies substantially between individuals based on gut microbiome composition.
This is the actual pharmacokinetic explanation for why two people taking identical 500mg NMN capsules from the same bottle report completely different outcomes. It is not placebo effect variability. It is microbial population variability.
Liposomal protection here: Microbiota exist in the gut lumen. Liposomal NMN’s endocytic pathway delivers intact NMN directly into intestinal epithelial cells — inside the cell wall, inaccessible to luminal bacteria. The deamidation threat is bypassed at the delivery level.
Does Standard Capsule NMN Still Work?
Yes — and this matters for honesty.
The major human RCTs (Yoshino 2021 in Science, Igarashi 2022 in npj Aging, Pencina 2023) all used standard capsule NMN and confirmed measurable tissue NAD+ elevation. Standard capsules clear the minimum efficacy threshold at 500mg/day from a verified manufacturer.
What standard capsules cannot do is match the bioavailability ceiling of a correctly specified liposomal product. If a standard capsule delivers 40–60% of label dose as intact intracellular NMN, and liposomal delivers 70–85%, the nominal cost premium for liposomal narrows significantly when you calculate cost per delivered milligram rather than label milligram.
The hierarchy, bluntly stated: Liposomal > Enteric-coated > High-quality standard capsule > Generic powder.
What a Legitimate Liposomal NMN Product Must Specify
Not all liposomal NMN products are what they claim. Four manufacturing specifications determine whether the liposomal mechanism actually works:
- Particle size: 50–200 nm — confirmed by dynamic light scattering (DLS). Above 500nm, endocytic efficiency collapses.
- Encapsulation efficiency: >85% — free NMN in a “liposomal” product faces the same three-layer attrition as a standard capsule.
- Phospholipid source: Sunflower lecithin — more stable bilayer architecture than soy-derived phospholipids; no allergen risk.
- Format: Powdered capsule over liquid — liposomes degrade faster in liquid suspension during shipping and storage. Powder format has substantially better shelf stability.
If a brand cannot disclose these four specifications, the liposomal claim is marketing, not chemistry.
The Bottom Line
NMN bioavailability is not one problem. It is three sequential, cumulative degradation threats: gastric hydrolysis at pH 1.5–3.5, CD73 brush border interception, and gut microbial deamidation. Standard capsule NMN survives all three imperfectly. Correctly specified liposomal NMN addresses all three mechanistically.
Before spending $60–$100/month on any NMN product, these are the questions worth asking your brand. The answers — or the absence of them — tell you everything.
