Elementary Idea About G-Proteins

G proteins are the family of proteins that have a significant role in cell signaling. “G” stands for “Guanine Nucleotide Binding Protein” which binds to its receptor G-protein Coupled Receptor (GPCR) to switch the cascade of signaling pathways. They are involved in transmitting signals from various receptors on the cell surface to the intracellular signaling pathways. These intracellular signaling pathways are responsible for regulating a wide range of cellular processes, including growth, metabolism, secretion, and gene expression.

The process by which G proteins transmit signals from receptors to the intracellular effectors is called signal transduction. G proteins are inactive in the GDP-bound state and active in the GTP-bound state. When an extracellular ligand (such as a hormone, gas, or neurotransmitter) binds to a specific receptor (GPCR) on the cell surface, it induces a conformational change in the receptor and in turn activates the associated G proteins.

Types of G-protein

G proteins can be classified broadly into two main types based on their size, structure, and complexity.

  1. Small G proteins

Small G proteins, also known as monomeric GTPases are single subunit proteins that act as a molecular switch. The general function of being active in the GTP-bound state and inactive in the GDP-bound state prevails to regulate the intracellular signaling pathways. These proteins are involved in several functions like cell growth, cytoskeletal, cell signaling, and trafficking.

Examples of small G proteins include:

· Ras — Important in the regulation of cell growth, differentiation, proliferation, and survival.

· Rho — Involved in cytoskeletal organization and cell migration.

· Rab — Involved in vesicle trafficking and membrane fusion.

· Arf — Helps in membrane trafficking and actin remodeling in Golgi bodies.

· Ran — helps in transportation in the nucleus and cytoplasm.

2. Large G proteins (Heterotrimeric)

Large G proteins also known as heterotrimeric G proteins are composed of three subunits: an alpha (α) subunit, a beta (β) subunit, and a gamma (γ) subunit. They bind to GPCR on the cell surface. The alpha subunit acts as a molecular switch in cycling between active and inactive states similar to the small G proteins but they differ in structure and mode of action.

Examples of G proteins include:

· Gαs — Gαs protein is involved in activating adenylyl cyclase, leading to an increase in cyclic adenosine monophosphate (cAMP) levels.

· Gαi — Gαi protein inhibits adenylyl cyclase, leading to a decrease in cAMP levels.

· Gαq — Gαq protein activates phospholipase C (PLC) leading to the production of inositol trisphosphate (IP3) and diacylglycerol (DAG).

· Gαt — Visual Signalling

Both small and large G proteins play a crucial role in cellular signaling and regulate a plethora of physiological processes. Their activation and activation cycles are tightly regulated and are important for many cellular responses. Thus it forms the central part of cellular communication and homeostasis.
