Multimodal Single-cell-resolved Spatial Proteomics

New Development Platform

Tissue cytometry is at the forefront of integrative analysis, transforming complex data into meaningful insights for scientists and patients alike. After significant effort, we have successfully combined tissue microdissection with mass spectrometry to create a multimodal, single-cell-resolved spatial proteomics platform.

One major challenge in proteomics is obtaining appropriate samples for mass spectrometry. Many clinical tumor biopsies contain only 10–50% tumor area, making it difficult for the resulting proteomic data to accurately represent the natural components of the tumor microenvironment. To address this challenge, we are collaborating with leading scientists and advanced instrument providers to launch this innovative platform.

The TissueFAXS cytometry system is used to image and analyze mIHC-labeled FFPE or fresh-frozen slides. By leveraging unique nuclei segmentation algorithms, deep neural networks, and AI classifiers, the system provides single-cell data. Users can perform quality control using intensity- and morphology-related gating, similar to size gating or fluorescence intensity cutoffs in flow cytometry. This process ensures that the data is derived from precisely defined cell populations.

The TissueFAXS cytometer goes beyond conventional imaging systems by offering precise image-to-data correlation. It allows the X and Y positions of target cells to be displayed on the slide and visualized within the images. These coordinates can then be exported to a laser microdissection system for the collection of specific target cells. Finally, the proteomics data is highly correlated with target cells through the purification of the sampling by TissueFAXS and SISPROT.

The Multimodal Single-cell-resolved Spatial Proteomics Platform was recently successfully installed at The Hong Kong University of Science and Technology. Researchers will use this platform to uncover the spatial complexities of the tumor microenvironment and neurodegenerative diseases.
